I found two methods of staining good. One suggested to me byProf. Henry Dixon, Professor of Botany, Trinity College, Dublin^viz. stain the dried and fixed culture witli 50 per cent. DelafielcVslogwood for ten minutes; then wash, dehydrate with alcohol, andmount in Canada balsam*: The other, the formic acid and borax methylene blue method, whichSabourand recommends for staining the hairs and scales of Fig. -2. Technique.—A drop of water is placed in the centre of a slide. Aminute amount of the culture is taken with a sterilised platinumneedle and transferred to the drop of water, and very gently movedabout in the water to dissociate it, if contact with the water does notimmediately lead to dissociation. Eough treatment would separatethe spores from the mycelium. The slide is then placed in an incubator till the water hasevaporated, or it may be allowed to diy in the air. The film is then fixed by placing over it a drop of formic acid, andthen heating the slide till the formic acid steams, letting it cool andheating again ; this process is repeated a few times.
