Each vessel contained 10 ml. of medium and was inoculated with ml. of a bacteria-free unialgal stock culture. Stock cultures from which the inocula were made are maintained at ° C. ± 1° C. in a light room. The basal medium consisted of the following: KH.,PO4, 20 mg.; thiamine HC1, mg.; biotin, 2 ; vitamin B-12, 2 /ig.; XaXO.,, 150 mg.; ferric sequestrene (13% Fe), 10 mg.;. FIGURE 1. Experimental temperatures were maintained in a series of constant temperature baths regulated to ± ° C. The culture flasks were submerged on plastic mesh racks into the water baths and incubated at temperatures ranging from 8° C. to 39° C.
