An important factor in the utilization of the Feulgen procedure for cyto- photometric purposes is the precise control of the hydrolysis step. When small pieces of tissue—rather than thin sections—are used, the hydrolysis procedure becomes subject to more variation. The end result is that DNA-Feulgen values for the same tissue processed in two separate procedures may be more variable.
